چکیده

Purpose: To investigate the potential role of miR-423-5p in osteoblastic differentiation non-traumatic osteonecrosis femoral head (ONFH).
Methods: MiR-423-5p levels bone marrow samples from ONFH and osteoarthritis (OA) patients, respectively, were evaluated using quantitative (real-time) polymerase chain reaction (qPCR). Osteoblastic was monitored Alizarin red S staining, while cell viability determined by MTT assay hMSC-BM. expression also measured during differentiation. The underlying mechanisms explored TargetScan database, a series vitro experiments performed to confirm this.
Results: significantly upregulated (p < 0.01) increased human mesenchymal stem cells-bone (hMSC-BM) after morphogenetic protein 2 (BMP-2) treatment. Furthermore, downregulated suppressed viability. SMAD3 observed be downstream target via bioinformatics analysis; further confirmed Conclusion: downregulates targeting osteonecrosis. Thus, may serve as for promoting patients.

MiR-708 promotes steroid-induced osteonecrosis of femoral head, suppresses osteogenic differentiation by targeting SMAD3.

Steroid-induced osteonecrosis of femoral head (ONFH) is a serious complication of glucocorticoid (GC) use. We investigated the differential expression of miRs in the mesenchymal stem cells (MSCs) of patients with ONFH, and aimed to explain the relationship between GC use and the development of MSC dysfunction in ONFH. Cells were collected from bone marrow of patients with ONFH. Samples were ass...

miR-424-5p promotes proliferation of gastric cancer by targeting Smad3 through TGF-β signaling pathway

MiRNAs have been reported to regulate gene expression and be associated with cancer progression. Recently, miR-424-5p was reported to play important role in a variety of tumors. However, the role and molecular mechanisms of miR-424-5p in GC (gastric cancer) remains largely unknown. In this study, we aimed to explore the role of miR-424-5p in GC. QRT-PCR was used to determine the expression leve...

Targeting miR-423-5p Reverses Exercise Training–Induced HCN4 Channel Remodeling and Sinus Bradycardia

RATIONALE Downregulation of the pacemaking ion channel, HCN4 (hyperpolarization-activated cyclic nucleotide gated channel 4), and the corresponding ionic current, If, underlies exercise training-induced sinus bradycardia in rodents. If this occurs in humans, it could explain the increased incidence of bradyarrhythmias in veteran athletes, and it will be important to understand the underlying pr...

MiR-490-5p Functions as an OncomiR in Breast Cancer by Targeting NFATc4

Breast cancer is a serious health problem worldwide in women. MicroRNAs are small non-coding RNAs of 18–25 nucleotides in length that post-transcriptionally modulate gene expression. MiR-490 has been reported as a tumor suppressor and oncomiR microRNA in breast cancer with two separate targets, NFAT and Rho. NFAT is one of the targets for miR-490 but the relationship between hsa</e...

Long non-coding RNA HOTAIR inhibits miR-17-5p to regulate osteogenic differentiation and proliferation in non-traumatic osteonecrosis of femoral head

BACKGROUND AND AIM The biological functions of non-coding RNAs (ncRNAs) have been widely identified in many human diseases. In the present study, the relationship between long non-coding RNA HOTAIR and microRNA-17-5p (miR-17-5p) and their roles in osteogenic differentiation and proliferation in non-traumatic osteonecrosis of femoral head (ONFH) were investigated. METHODS The expression levels...
